Any thoughts on the best route to drive? I see two that are only about 20 minutes difference.

Generally I find it is more about roads than distance. An established toll road is best, a libre road often is not so good but its a gamble.
Most seem to use route 5. going to San Felipe, in fact that is the route San Felipe recommends.

Thanks, that was my thought also. Not doing it anytime soon, but will in the not to distance future.
